The aquaporin (AQP) proteins facilitate the transport of water and small neutral solutes across cell membranes. AQP11 is expressed in brain tissue and at the blood-brain barrier (BBB) in mice, where it plays an important role in preventing brain edema. However, its exact function is unclear.
Order AQP11 antibody mutation that introduces a cysteine-to-serine substitution at amino acid 227 is known to cause severe polycystic kidney disease in humans and mice. To address this issue, we studied the relationship between AQP11 insufficiency and glucose-induced oxidative stress in kidney tubules (PT). We found that AQP11 expression was enhanced in the S1 segment of PTs, which is the major site for reabsorption and transport of glucose along the nephron. Moreover, we demonstrated that glucose-induced AQP11 expression is associated with the activation of caspase-3, which is a downstream executor of ER stress and mitochondrial dysfunction.
Order AQP11 Antibody: Where to Get Reliable Products
To analyze the oligomeric structure of AQP11, we performed native PAGE analyses using kidney cortex tissues from wild-type and Aqp11sjds/sjds mutant mice. In the homozygous mutant, the lower molecular mass form of AQP11 is significantly decreased and an additional intermediate band corresponding to different oligomeric forms is present. This indicates that the loss of AQP11 in Aqp11sjds/sjds mice results in reduced formation of the hydrophilic oligomeric form of this protein, which is necessary for its function. Furthermore, we confirmed that the cysteine-to-serine modification of AQP11 affects its oligomeric stability. Moreover, we showed that the oligomeric form of AQP11 is associated with its resistance to glucose-induced oxidative stress in PTs.
